India has approved its first dengue vaccine, QDENGA, following clearance by the Drug Controller General of India (DCGI). The vaccine, developed by Takeda Biopharmaceuticals India, is intended for individuals aged 4 to 60 years and can be administered regardless of prior dengue infection. Unlike some other dengue vaccines, QDENGA does not require pre-vaccination screening, making it accessible to a broader population without the need for prior testing. The approval comes after extensive clinical trials and long-term data analysis. The vaccine is part of Takeda's global clinical development program, which included 19 phases of trials involving over 28,000 participants in both dengue-endemic and non-endemic regions. The Phase III DEN-302 trial conducted specifically in India evaluated the vaccine's safety and immune response among individuals aged 4 to 60. The results contributed significantly to the regulatory decision. QDENGA is a tetravalent, live-attenuated vaccine designed to offer protection against all four dengue virus serotypes. It is administered as two subcutaneous injections of 0.5 ml, spaced three months apart. The vaccine has already been approved in 43 countries, with more than 32 million doses distributed globally. Takeda reports that the latest seven-year data confirm the vaccine's ongoing effectiveness in preventing dengue infection and hospitalization across all four serotypes. India bears a substantial portion of the global dengue burden, accounting for nearly one-third of all cases. According to the company, the actual number of infections could be much higher than officially reported, with estimates suggesting tens of millions of infections occur annually. Factors contributing to this include rapid urbanization, climate change, and widespread mosquito breeding. These conditions have turned dengue into a year-round public health issue, placing millions of Indians at risk each year. The World Health Organization (WHO) recommends QDENGA for use in dengue-endemic regions and supports its integration into public immunization programs in high-transmission areas without requiring pre-vaccination screening. The vaccine has also received WHO prequalification, ensuring it meets international standards for quality, safety, and efficacy. This status allows QDENGA to be procured by global agencies such as UNICEF and the Pan American Health Organization (PAHO), enhancing its availability in low-resource settings. Takeda's leadership emphasized the significance of this approval in strengthening India's approach to dengue prevention. Dr. Mahender Nayak, Head of Intercontinental Markets, noted that the vaccine's approval marks a critical step in combating dengue, particularly in a country where the disease remains a persistent challenge. Similarly, Dr. Goh Choo Beng highlighted the importance of combining vaccination with vector control, surveillance, and community awareness to address the multifaceted nature of dengue outbreaks. With QDENGA now available, the focus will shift towards ensuring equitable distribution and accessibility. Public health officials and Takeda are likely to collaborate on strategies to educate the public about the vaccine, monitor its impact, and integrate it into existing healthcare frameworks. As the vaccine becomes widely available, it is expected to play a key role in reducing the incidence and severity of dengue infections in India.
