Scientists may have uncovered a natural defense mechanism against aging, though the potential benefits come with caveats. A recent study published in Nature Communications reveals that a hormone called thymulin, which declines with age, plays a critical role in controlling inflammation, a factor strongly associated with age-related diseases such as cancer, heart disease, and neurodegeneration. Researchers at the Keck School of Medicine of USC discovered that restoring thymulin levels in older mice significantly reduced inflammation and enhanced their ability to combat cancer. The study focused on the thymus, a small gland located behind the breastbone responsible for training the immune system. While the thymus is well-known for producing T-cells, the researchers identified another function: thymulin, a hormone secreted by the thymus, helps regulate inflammatory responses by managing cytokine activity. Cytokines are signaling molecules that can either promote or suppress inflammation. As thymulin levels decrease with age, cytokine production increases, leading to a state of chronic inflammation known as inflammaging. To investigate this phenomenon, scientists employed a unique method involving pairing older and younger mice to share a common circulatory system. This allowed them to observe how factors present in young blood affected aging processes. The results indicated that exposure to young blood reduced inflammation in older mice, suggesting that thymulin, or similar substances, might hold therapeutic promise. Further analysis of human blood samples and mouse models confirmed the link between thymulin and aging, reinforcing its role in maintaining immune balance. The implications extend beyond inflammation. In experiments with older mice suffering from cancer, administering thymulin led to stronger immune responses against tumors and improved survival rates. Moreover, the hormone enhanced the efficacy of anti-PD-1/PD-L1 immunotherapies, which are commonly used to treat certain cancers. These findings highlight the possibility that age-related decline in thymulin contributes not only to chronic inflammation but also to diminished immune function in later years. Despite these promising results, the study was conducted exclusively in mice, and there is currently no evidence that thymulin can prevent or cure cancer in humans. Researchers emphasize that while the discovery identifies a potential biological pathway worth exploring, much work remains before clinical applications can be considered. Dr. Fumito Ito, the lead author of the study, noted that the research offers the first evidence that a naturally occurring substance from the thymus declines with age and may reverse age-related inflammation. Looking ahead, scientists aim to determine whether thymulin influences tumor formation and progression, as well as its potential role in other age-related conditions such as diabetes, chronic kidney disease, and Alzheimer’s. Prof. Ercüment Ovalı, a hematologist and stem cell biologist unaffiliated with the study, acknowledged the significance of the findings but stressed that targeting immune aging remains in the early stages of research. He emphasized that while such approaches could eventually aid in treating age-related diseases, especially cancer, they require extensive testing before becoming viable medical interventions. Cancer treatment outcomes, according to Ovalı, depend on more than just the tumor and the drugs used. The biological condition of a patient’s immune system plays a crucial role in determining the success of therapies. As the field continues to evolve, the search for ways to harness the body’s natural defenses against aging and disease will likely remain a central focus of biomedical research.
